Prokaryotic DNA-binding proteins, H-NS and HU function as regulators of gene expression. The HU protein is a small, basic, heat-stable DNA-binding protein that is well-conserved in prokaryotes and is associated with the bacterial nucleoid. Hu protein not only functions to bind DNA but also binds to RNA and DNA-RNA hybrids with the same affinity as supercoiled DNA. H-NS proteins plays a role in the formation of nucleoid structure and affect gene expression under certain conditions.
