ColE1 replication: What would happen if you over-expressed the first 600 base of RNA II by introducing a truncated copy of the RNA II primer gene into the plasmid? For example, the ColE1 plasmid would have a second copy of the RNA II gene (including a strong promoter) that would only be comprised of nucleotides 1-600.

    It will lead to increase in DNA replication with a corresponding increase in copies of the plasmid. This is because the over expression will enable RNA II to avoid RNA 1 disruptions; now with a corresponding increase in the primers (introduced), priming increases with consequent increase in DNA synthesis.
