You are working to characterize a novel protein in mice. Analysis shows that high levels of the primary transcript that codes for this protein are found in tissue from the brain, muscle, liver, and pancreas. However, an antibody that recognizes the C-terminal portion of the protein indicates that the protein is present in brain, muscle, and liver, but not in the pancreas. What is the most likely explanation for this result?

    Firstly you have to reconfirm the results by repeating the experiments and all the reagent and solutions are totally fine and also assure that there is no human error.

    So is transcript is present, so after post transcription modification there could be some changes occurred in the sequence of protein. for me the reason is that during alternative splicing, some part of the transcript is removed which encode the binding region for antibody binding. so when the protein is translated, it will miss the region C-terminal for antibody binding and could not be detected.
