You have two tubes (I and 2) that initially contain the same concentration of starch. However, you add one microgram (1ug) of amylase enzyme to tube 2 and then let both tubes sit for ten minutes. The resulting absorbance value for tube 1 is 0.74 and for tube 2 is 0.56. why do you see a difference between the two samples?

The initial concentration is 10 because it should be the same as tube A which lacks the enzyme breaking down the starch. And the final concentration is 5. The concentrations come from the correspondence with the absorbance on the graph
