The significance of capitalizing the word "Posterity" in the Preamble is to show it's importance. Posterity means "All future generations of people." For them to capitalize it is huge because they want it to leave a legacy behind. They want everyone to know how important the future generations are to them so they capitalized it to imply the importance. "In order to form a more perfect union, establish Justice, insure domestic Tranquility" is implying that they want the future generation to live peaceful, freely and happy.
